Composition

Pharmacological action
Oestriol is an oestrogenic compound and binds to the oestrogen receptors. Unlike other oestrogens (eg. oestradiol and its esters and ethinyloestradiol) the nuclear occupance time of oestriol is short. Oestriol is therefore a short acting oestrogen.

In the incontinent female dog it has a beneficial effect on urinary incontinence. Upon oral administration a steady state is reached after the seconded treatment and no accumulation occurs after multiple dosing. Urinary incontinence is described as an involuntary leakage of urine. While there are a number of causes of urinary incontinence in the bitch urethral sphincter mechanism incompetence (SMI), which usually occurs after ovariectomy, is most common.

As the urethral sphincter contains oestrogen receptors and oestrogens appear to be effective in the treatment of urinary incontinence, it has been strongly suggested that there is a relationship between the incidence of sphincter mechanism incompetence (SMI) and oestrogen depletion.

Indications
Incurin is indicated for the treatment of hormone-dependent urinary incontinence due to sphincter mechanism incompetence (SMI) in ovariohysterectomised bitches.

Contra-indications

  • Intact bitches
  • Pregnancy
  • Animals with diabetes.
  • Animals showing a polyuria-polydipsia syndrome should not be treated with Incurin.
  • Dogs younger than 1 year of age.

Warnings
High doses of oestrogen may have a tumor-promoting effect in target organs with oestrogen receptors (mammary gland).

Dosage and Directions for Use

  • Administer orally once daily.
  • Since no relation exists between the final effective dose and body weight, a fixed dose per kg body weight is not feasible. The dose has to be determined for each dog on an individual basis.

The following dosing schedule is suggested:

  • Start the treatment with 1 tablet every day.
  • If the treatment is successful, lower the dose to half a tablet per day. If the treatment is not successful, increase the dose to 2 tablets per day.
    Some dogs do not need daily treatment; treatment every other day may be tried once an effective dose has been established. The minimum dose given should not be less than 0.5mg per dog per day. Ensure the dose used to achieve the therapeutic effect is as low as possible. Do not use more than 2 tablets per dog per day. If no response to treatment is obtained the diagnosis should be reconsidered in order to investigate other causes for the incontinence such as neurological disorders, bladder neoplasia, etc. Animals should be re-examined every 6 months during treatment.

Side-effects and Special Precautions

  • Mild oestrogenic effects such as a swollen vulva, swollen mammae and/or attraction for males at the high dose of 2 mg have been observed. These effects are reversible after lowering the dose.
  • Symptoms of nausea have been observed.

White to almost white, round, flat, scored tablets.

Presentation
Blister pack of PVC/aluminium foil containing 30 x 1 mg tablets per strip. The strips are packed in cardboard boxes.

Storage Instructions
Store at room temperature below 25 ºC. Keep out of reach of children and uninformed persons.
